XORTX Therapeutics Advances XORLO for Gout with FDA Submission on the Horizon

Gout, a painful form of inflammatory arthritis, is set to become more prevalent in the coming decades. Projections indicate that the number of cases globally will rise significantly over the next 25 years, driven by an aging population, rising obesity rates, and lifestyle factors. In the United States alone, over 9 million people have been diagnosed with gout, and an even larger group—44 million people—have elevated uric acid levels, placing them at risk for the disease. This growing burden underscores the need for better treatment options.

Current therapies, primarily oral xanthine oxidase inhibitors (XOIs), are effective but come with significant drawbacks. Some patients experience severe gastrointestinal issues, kidney toxicity, and cardiovascular risks, especially with long-term or high-dose use. Moreover, 3% to 5% of patients cannot tolerate these drugs, and they carry boxed warnings for cardiovascular risk. These limitations often lead to poor adherence and inadequate disease management.

XORTX Therapeutics Inc. (NASDAQ: XRTX), a late-stage clinical pharmaceutical company, is working to change that. The company has developed XORLO™, a proprietary formulation of oxypurinol that utilizes a liposomal delivery system to enhance oral absorption. This allows for lower total daily doses while effectively lowering uric acid levels. XORLO is designed to overcome the poor oral absorption of oxypurinol, minimizing toxic exposure and severe gastrointestinal side effects associated with traditional high-dose therapies. According to XORTX, this differentiated approach could offer a better-tolerated option for gout sufferers.

XORTX is making concrete progress toward bringing XORLO to market. The company recently announcing the launch of drug material and tablet manufacturing to support a planned two-part clinical study, XRX-OXY-102. This study is based on positive prior clinical results and discussions with the FDA. The production of pure oxypurinol drug substance using XORTX's proprietary synthetic pathway has already begun, and the company is working with a contract manufacturing partner to produce commercial drug batches. The clinical trial will characterize the pharmacokinetics and efficacy of XORLO, and the resulting data will support a New Drug Application (NDA) submission.

The company is also working toward a new drug application submission to the FDA, with a planned IND submission in the fourth quarter of 2026. If the two-part clinical trial is successful, XORTX intends to request a pre-NDA meeting and prepare an NDA submission while engaging in pre-commercialization activities. The company is targeting an NDA submission in about one year and estimates a market opportunity of $700 million annually for XORLO. "Our planned IND submission to the FDA next quarter is a defining step for the XRx-026 program," said Allen Davidoff, Co-CEO of XORTX. "It reflects the strength of our prior clinical results and moves us closer to potentially offering gout patients a differentiated treatment option."

The implications of XORTX's progress are significant. With gout prevalence on the rise, there is a pressing need for therapies that are both effective and well-tolerated. XORLO's potential to provide rapid uric acid reduction with lower dosing could improve patient adherence and outcomes. If approved, it would offer a new option for the millions of people who cannot tolerate or do not respond adequately to existing treatments. As XORTX advances toward an NDA, it is positioning itself to address a substantial unmet need in gout care. For more information, the original release is available on www.newmediawire.com.
